Whole Genome Profiling

High quality Bacterial Artificial Chromosome (BAC) libraries form an important basis in KeyGene's genetic and genomic research. KeyGene has constructed a wide range of BAC libraries for own use and for external parties.

Benefits

  • Full service – including BAC library construction, BAC pooling, Whole Genome Profiling, contig assembly and customer support;
  • Pricing – comparable or more cost effective than BAC end Sequencing or SnapShot technology;
  • WGP generates 30 bp sequence tags that are spaced every ~2 Kb to 3 Kb across the BAC clones, providing unparalleled quality in contig building;
  • WGP provides an excellent framework for the assembly of entire genomes - Whole genome Sequence information can be placed on the framework to establish a high quality genome sequence;
  • WGP marker data can be added immediately to an existing genome assembly;
  • BAC clones are positioned in contigs across the genome - gene cloning or re-sequencing projects in regions of interested are greatly facilitated by this.

Deliverables

  • The deliverables will be a data carrier with all the WGP data and customized FPC software to visualize and edit the WGP maker/sequence data as well as living BAC clones on 384 well format if a custom BAC library was made.

Examples

  • KeyGene and Amplicon Express performed a "test" with WGP on Arabidopisis (~135 Mbp genome);
  • Melon (450 Mbp genome) - 550 contigs at very high stringency;
  • Tomato Genome (950 Mb) physical map;
  • a 2,600 Mbp plant genome project was started with very good initial results. Project is ongoing.
